Analysis of 45 sera was performed employing five techniques which are currently in use in three laboratories to measure anti-Pasteurella haemolytica antibodies. The enzyme linked immunosorbent assay, passive hemagglutination, complement fixation and direct and indirect bacterial agglutination assays were employed and a relationship between tests in the measurement of anti-P. haemolytica antibodies was demonstrated. Regression analysis together with prediction and confidence intervals were tabulated also. The conclusion drawn from statistical analysis was that all five tests are similar in their ability to detect immune responses (antibody and antigen(s) interactions) to Pasteurella haemolytica.  相似文献

Designing fishing policies without knowledge of past levels of target species abundance is a dangerous omission for fisheries management. However, as fisheries monitoring started long after exploitation of many species began, this is a difficult issue to address. Here we show how the ‘shifting baseline’ syndrome can affect the stock assessment of a vulnerable species by masking real population trends and thereby put marine animals at serious risk. Current fishery data suggest that landings of the large Gulf grouper (Mycteroperca jordani, Serranidae) are increasing in the Gulf of California. However, reviews of historical evidence, naturalists’ observations and a systematic documentation of fishers’ perceptions of trends in the abundance of this species indicate that it has dramatically declined. The heyday for the Gulf grouper fishery occurred prior to the 1970s, after which abundance dropped rapidly, probably falling to a few percent of former numbers. This decline happened long before fishery statistics were formally developed. We use the case of the Gulf grouper to illustrate how other vulnerable tropical and semi‐tropical fish and shellfish species around the world may be facing the same fate as the Gulf grouper. In accordance with other recent studies, we recommend using historical tools as part of a broad data‐gathering approach to assess the conservation status of marine species that are vulnerable to over‐exploitation.  相似文献

  1. The ocean crisis is urgent and central to human wellbeing and life on Earth; past and current activities are damaging the planet's main life support system for future generations. We are witnessing an increase in ocean heat, disturbance, acidification, bio‐invasions and nutrients, and reducing oxygen levels. Several of these act like ratchets: once detrimental or negative changes have occurred, they may lock in place and may not be reversible, especially at gross ecological and ocean process scales.
  2. Each change may represent a loss to humanity of resources, ecosystem function, oxygen production and species. The longer we pursue unsuitable actions, the more we close the path to recovery and better ocean health and greater benefits for humanity in the future.
  3. We stand at a critical juncture and have identified eight priority issues that need to be addressed in unison to help avert a potential ecological disaster in the global ocean. They form a purposely ambitious agenda for global governance and are aimed at informing decision‐makers at a high level. They should also be of interest to the general public.
  4. Of all the themes, the highest priority is to rigorously address global warming and limit surface temperature rise to 1.5°C by 2100, as warming is the pre‐eminent factor driving change in the ocean. The other themes are establishing a robust and comprehensive High Seas Treaty, enforcing existing standards for Marine Protected Areas and expanding their coverage, especially in terms of high levels of protection, adopting a precautionary pause on deep‐sea mining, ending overfishing and destructive fishing practices, radically reducing marine pollution, putting in place a financing mechanism for ocean management and protection, and lastly, scaling up science/data gathering and facilitating data sharing.
  5. By implementing all eight measures in unison, as a coordinated strategy, we can build resilience to climate change, help sustain fisheries productivity, particularly for low‐income countries dependent on fisheries, protect coasts (e.g. via soft‐engineering/habitat‐based approaches), promote mitigation (e.g. carbon storage) and enable improved adaptation to rapid global change.

Analysis of published CD5 amino acid sequences identified conserved sequences with potentially immunogenic epitopes. To obtain anti-porcine CD5, synthetic peptides representing conserved sequences identified in mouse, human, cattle and sheep CD5 cytoplasmic tail domains were linked to KLH and used to immunize rabbits. Anti-synthetic peptide serum reacted with an antigen extracted from porcine lymphocyte membrane which was consistent in size (67 kDa) with CD5. Murine monoclonal anti-porcine wCD5 (b53b7) and the anti-CD5 synthetic peptide serum react with the same ligand confirming that porcine wCD5 has conserved amino acid sequences similar to those of CD5 of several species. Analysis of porcine genome for CD5 gene sequences by PCR was conducted to verify the presence of CD5-like genes. Oligomeric primers were designed to identify CD5-like sequences by polymerase chain reaction in pigs and other species. Amplified DNA similar in size to that predicted for CD5 elements were amplified from a variety of animal genomes including that of pig. The porcine-derived fragment was cloned and shown to be 96% similar to mouse CD5. The use of published CD sequences for prediction of immunogenic peptides has provided a complimentary alternative to the more traditional approaches to production of CD-specific antibodies.  相似文献

Forsyth, L.M.G., Jackson, L.A., Wilkie, G., Sanderson, A., Brown, C.G.D. and Preston, P.M., 1997. Bovine cells infected in vivo with Theileria annulata express CD11b, the C3bi complement receptor. Veterinary Research Communications, 21 (4), 249-263Bovine cells from cattle infected with Theileria annulata were phenotyped with monoclonal antibodies recognizing bovine leukocyte antigens. Macroschizont-infected, transformed cell lines prepared from peripheral blood mononuclear cells of cattle, infected with sporozoites, were assessed by flow cytometry; parasitized cells in tissues from infected cattle were examined by immunocytochemical techniques. Co-expression of markers for different cell lineages by the cell lines precluded a definite conclusion as to their phenotypic origins. For, while the pattern of leukocyte antigens expressed by these in vivo-derived schizont-infected cells, which included CD11b, was indicative of a myeloid origin, the possibility that they were NK cells could not be excluded. The monoclonal antibody (MAb) IL-A15, which recognizes CD11b, reacted with a high proportion of parasitized cells in sections of tissues from infected cattle at all stages of acute disease. Mononuclear cells infected with parasites at all stages of differentiation, from macroschizont to microschizont, expressed CD11b. Such parasitized cells occurred throughout the lymphoid tissues, being found in the thymus, spleen and lymph nodes, particularly the prescapular node draining the site of infection, the hepatic, mesenteric and precrural nodes, as well as in the reticulo-endothelial tissue of the liver, kidney, lung, abomasum, adrenal and pituitary glands. These observations provided the first evidence for a myeloid origin for the parasitized T. annulata cells found in infected bovine tissues and blood and suggested a mechanism whereby schizonts could transfer from cell to cell during mechanical infection with schizont-infected cells.  相似文献

This study examines factors affecting the rate and extent of biomass build-up among commercially important groupers, snappers, grunts, parrotfish and surgeonfish in a network of four marine reserves in southwest St. Lucia, Caribbean. Reserves constituted 35% of the total reef area originally available for fishing. Protection was instigated in 1995 after a baseline survey with annual or biennial censuses performed until 2002. Each survey consisted of 114 fifteen minute fish counts in reserves and 83 in fishing grounds, at depths of 5 m and 15 m in a 10 m diameter counting area. Estimates of number and size (cm) of target species were used to calculate fish family biomass. Data were analysed using three-way ANOVA in a before-after-control-impact pairs (BACIP) design. All families increased significantly in biomass over time at nearly all sites. Increases were greater in reserves than fishing grounds, except for grunts, and responses were strongest in parrotfish and surgeonfish. The combined biomass of families more than quadrupled in reserves and tripled in fishing grounds between 1995 and 2002. During this period coral cover declined by 46% in reserves and 35% in fishing grounds. Multiple regression showed that neither habitat characteristics nor habitat deterioration significantly affected rates of biomass build-up. The key factor was protection from fishing, which explained 44% of the variance in biomass growth. A further 28% of the variance was explained by sedimentation, a process known to stress reef invertebrates, significantly reducing the rate of biomass build-up. St. Lucia’s reserves succeeded in producing significant gains to fish stocks despite coral cover and structural complexity falling steeply over the period of the study.  相似文献

Adult and juvenile mobility has a considerable influence on the functioning of marine protected areas. It is recognized that adult and juvenile movement reduces the core benefits of protected areas, namely protecting the full age–structure of marine populations, while at the same time perhaps improving fisheries yield over the no-reserve situation through export of individuals from protected areas. Nevertheless, the study of the consequences of movement on protected area functioning is unbalanced. Significant attention has been paid to the influence of certain movement patterns, such as diffusive movement and home ranges, while the impacts of others, such as density-dependent movements and ontogenetic migrations, have been relatively ignored. Here we review the diversity of density-independent and density-dependent movement patterns, as well as what is currently known about their consequences for the conservation and fisheries effects of marine protected areas. We highlight a number of ‘partially addressed’ issues in marine protected area research, such as the effects of reserves targeting specific life phases, and a number of essentially unstudied issues, such as density-dependent movements, nomadism, ontogenetic migrations, behavioral polymorphism and ‘dynamic’ reserves that adjust location as a realtime response to habitat changes. Assessing these issues will be essential to creating effective marine protected area networks for mobile species and accurately assessing reserve impacts on these species.  相似文献

Effects of topical administration of a single dose of 2% pilocarpine on intraocular pressure (IOP) and pupil diameter were evaluated in normotensive eyes of 10 clinically normal cats over 12 hours. Mean (+/- SEM) normal IOP was 17.1 (+/- 1.1) mm of Hg and, diurnal fluctuation was observed, with the highest IOP seen in the evening. Mean (+/- SEM) normal pupil diameter was found to be 10.1 (+/- 0.5) mm. Topical treatment with pilocarpine resulted in reduction of IOP in treated and nontreated eyes. This effect was time-dependent and was first observed at 4 hours after treatment. Mean reduction of IOP was 15.2% in the treated eye and 9.3% in the nontreated eye. The treated eye had reduced pupil diameter at 30 minutes after treatment, and miosis persisted throughout the 12 hours of the study. Mean reduction in pupil diameter was 28.5% in the treated eye and 14.2% in the nontreated eye. Topically administered pilocarpine results in reduction of IOP and pupil diameter in treated and contralateral eyes, which supports the use of pilocarpine for treatment of glaucoma in cats.  相似文献
